摘 要:目的 :研究归芪方对糖尿病大鼠肾脏病变的保护作用及其机制。方法 :Wistar大鼠腹腔注射链脲佐菌素(STZ)以建立糖尿病模型 ,随机分为糖尿病模型组 (D组 )、苯那普利治疗组 (B组 )、归芪方治疗组 (G组 ) ,另设正常对照组 (C组 )。实验第 4 ,6周时 ,观察各组大鼠肾重 /体重、血糖、内生肌酐清除率 (Ccr)及肾组织结构的改变 ,放免法测定 2 4h尿微量白蛋白排泄率 (UAER)、β2 -微球蛋白 (β2 -MG)、血浆及肾组织血管紧张素Ⅱ (AngⅡ )含量 ,原位杂交法检测肾皮质转化生长因子 - β1(TGF - β1)基因表达的变化。结果 :与糖尿病模型组相比 ,归芪方可控制肾脏肥大 ,改善肾功能 ,降低AngⅡ水平 ,使肾皮质TGF - β1mRNA表达量减少 ,同时肾脏病理改变亦减轻。结论 :归芪方可降低AngⅡ水平 ,抑制TGF - β1mRNA的表达 ,减轻肾脏损伤 ,改善肾功能 ,延缓糖尿病肾病的慢性病理进展。Objective: To investigate the protective mechanism of Guiqi Mixture(GQM),a traditional Chinese medicine, on the kidney of diabetic rats. Methods: The diabetic animal model was made by a single intraperitoneal injection of 60 mg·kg -1 streptozotocin(STZ). The rats were randomly assigned to four groups: diabetic control group(D),Benazepril-treated group(B), Guiqi Mixture-treated group(G) and normal control group(C). After 4 and 6 weeks of treatment,renal weight/body weight ratio,urine albumine excretion ratio(UAER) and creatinine clearance(Ccr) were measured.The renal pathological lesions were also investigated. The expression of renal transforming growth factor-β 1(TGF-β 1) mRNA in renal was determined by in situ hybridization and angiotensinⅡ(AngⅡ) concentration in plasma and kidney was investigated by radioimmunoassay. Results:UAER,β 2-MG, Ccr, renal weight/body weight ratio, plasma and kidney AngⅡ concentration of group G were decreased compared with those of group D.It could also significantly decrease the expression of TGF-β 1 mRNA. Conclusion: GQM may alleviate kidney lesion and slow down the progression of diabetic nephropathy by inhabiting the expression of TGF-β 1 mRNA and decrease the level of AngⅡ.
