摘  要:目的 探讨美白化学物熊果苷对皮肤黑素细胞的生物学效应。方法 利用B - 16黑素瘤细胞株 ,添加熊果苷 5d后 ,采用苔盼蓝排斥试验测定细胞生长情况 ,采用丙酮酸比色法测定细胞培养上清液中的乳酸脱氢酶 (LDH)释放水平 ,对熊果苷的细胞生物学效应进行评价 ,以RPMI 16 4 0培养液培养同期B - 16黑素细胞作正常对照。结果 加入不同剂量(10、30、5 0、70、10 0 μg/ml)熊果苷后 ,可使LDH明显升高 ,细胞增殖抑制率显著增强 ,黑素合成减少 ,但对酪氨酸酶活性抑制不明显。结论 熊果苷具有明显的细胞毒性 ,可抑制细胞增殖和黑素合成 ,其美白机制与酪氨酸酶活性改变的关系不大 。Objective This study is to investigate the bio-efficacy of arbutin on melanocytes in skin. Methods After B-16 melanoma cells were treated with arbutin for five days, pyruvic acid colorimetry was used to determine the release of cytoplasmic enzyme(Lactate dehydrogenase, LDH), trypan blue exclusion test was used to determine the condition of cell growth. Results After being treated with arbutin at doses of 10,30,50,70,100μg/ml respectively, the release of cultured cytoplasmic enzyme (LDH) increased, the inhibitory rate of cell proliferation increased, and the melanin content decreased significantly. The change of activity of tyrosinase was not significant. Conclusion The results showed that Arbutin is of marked cytotoxicity, inhibits the proliferation of cell and the melanin synthesis seriously, its lightening mechanism may relate to damage of the cell membrane, the inhibition of the melanin synthesis and, may not relate to the inhibition of tyrosinase activity
