摘  要:目的探讨人类转化生长因子β诱导基因-克隆3(TGF-β-inducedgene-human,clone3,βig-h3)在糖尿病大鼠肾组织中的表达意义,以及血管紧张素受体拮抗剂洛沙坦losartan和中药黄芪对糖尿病大鼠肾组织βig-h3表达的影响。方法采用链脲佐菌素(STZ)糖尿病大鼠模型。将SD大鼠随机分成5组,每组6只:正常对照组(N组)、糖尿病组(D组)、losartan治疗组(DL组)、黄芪治疗组(DA组)和losartan黄芪联合治疗组(DLA组)。治疗8周后取肾组织,观察肾脏病理改变;半定量RT-PCR法检测各组肾组织中βig-h3和TGF-β1mRNA的表达;免疫组化方法检测各组肾组织中βig-h3和TGF-β1蛋白质的表达水平。结果处理8周后,与N组相比,D组肾小球系膜增生、基质大量堆积,部分肾小球出现硬化;各治疗组系膜增生均有减轻。D组肾组织βig-h3、TGF-β1mRNA和蛋白表达均明显升高(与C组比较,P<0.05);各治疗组上述指标均有不同程度的下降(与D组相比,P<0.05)。βig-h3的表达与TGF-β1呈正相关(r=0.81,P<0.05)。结论βig-h3在糖尿病肾病发生发展过程中可能起重要作用;losartan和黄芪可能部分通过下调肾组织βig-h3mRNA和蛋白的表达对糖尿病肾病发挥抗纤维化作用。Objective To study the expression of βig h3,a newly found molecule of ECM,in the renal tissue of diabetic nephropathy and the significance of renal protection with losartan and Astragalus memberaneous.Methods STZ induced diabetic rats were used in this study.The expression of βig h3 and TGF β1 in the kidneys was assessed by RT PCR and immunohistochemistry, respectively, after eight weeks of treatment.Results The levels of βig h3 and TGF β1 mRNA in the diabetes group increased remarkably compared with those in the normal group(P< 0 05). In comparison with diabetes group, the levels of βig h3 and TGF β1 mRNA were significantly reduced by losartan,Astragalus memberaneous and combination of both(P< 0 05). The extent of βig h3 expression was positively correlated with the expression of TGF β1(r=0 81,P< 0 05).Conclusions βig h3 plays an important role in the development of diabetic nephropathy. Losartan and Astragalus memberaneous can exert a renal protection on diabetic nephropathy, possibly partly through the reduction of TGF β1 and βig h3 expression.
