补髓生血颗粒对慢性再障患者细胞凋亡及Bax基因的影响

摘　　要：目的 :探讨补髓生血颗粒对慢性再生障碍性贫血患者骨髓造血细胞过度凋亡和Bax基因表达水平的影响及其与中医辨证分型的关系。方法 :应用原位末端标记法及原位杂交法检测慢性再障患者应用补髓生血颗粒治疗前后骨髓造血细胞凋亡阳性指数及Bax基因表达阳性指数。结果显示 :治疗后慢性再障患者骨髓造血细胞凋亡阳性指数及Bax基因表达水平均下降 ,且阳虚型细胞凋亡阳性指数接近于正常 ,阴虚型与正常组比仍有较显著差异 ;而治疗后两型Bax水平与正常对照组比仍有较大差异。结论 :补髓生血颗粒对慢性再障患者骨髓造血细胞过度凋亡有抑制作用 ,下调Bax基因表达水平仅是其抑制细胞凋亡的机制之一。Purpose:To study the influence of Marrow-Supplementing and Blood-Engendering Granule to the excessive apoptosis and the Bax gene expression of chronic aplastic anemia(CAA)patients'hemopoietic cell.Methords:Use the technique of in situ end-labeling and in situ hybridization to examine the positive index of apoptotic cell and Baxgene expression of hemopoietic cell of CAA patients'bone marrow before and after the treatment with Marrow-Supplementing and Blood-Engendering Granule.Result:The positive index of the apoptotic cell and Bax gene expression of hemopoietic cell of CAA patients both decreased after the treatment.The positive index of the apoptotic cell of the group of kidney yang deficiency is close to to the normal group and that of kidney yin deficiency is much higher than the normal group.The positive index of Bax gene expression of both the group. of kidney yang deficiency and kidney yin deficiency is much higher than the normal group.Conclusion:Marrow-Supplementing and Blood-Engendering Granule can inhibit the excessive apoptosis of and Bax gene expression of hemopoietic cell of CAA patients.Inhibiting the Bax gene expression is only one of the mechanisms of inhibiting the excessive apoptosis.
