摘  要:目的 考察影响二苯乙烯苷 (THSG)溶液稳定性的因素 ,并计算室温下其晶体粉末的 t0 .9值。方法 采用HPL C法和 HPL C- MS联用法。结果 以 L ichrospher5 - C1 8(2 5 0 m m× 4 .6 m m,5 μm)为固定相 ,甲醇 -水 (4∶ 6 )为流动相 ,检测波长为 310 nm。 THSG进样量在 0 .14~ 2 .80μg与峰面积线性关系良好 ,自制 THSG纯度高达99.94 % ,p H5 .8时 ,THSG与水分子发生加成反应。强酸促使 THSG水解为葡萄糖和苷元 ,并进一步降解为酚类化合物。强碱性条件下 ,THSG氧化为醌类化合物。在中性及弱碱性水溶液中较稳定 ,30 d内含量无明显变化。THSG晶体粉末室温下放置 2 4 0 d含量无明显变化 ,t0 .9值为 10 98d。Object To study the factors which influenced the stability of stilbene glucosidel (tetrahydroxystilbene-glucoside, THSG) solution, investigate and calculate the value of t 0.9 of THSG crystal powder at room temperature. Methods HPLC and HPLC-MS methods were used. Results The best chromatographic conditions were to employ Lichrospher 5-C 18(250 mm×4.6 mm,5 μm) as the stable phase, methanol-water (4∶6) as mobile phase, 310 nm as the detective wavelength. Within the range from 0.14 to 2.80 μg, there was a good linearity. The content of self-prepared THSG was up to 99.94%. Stability test showed that when pH was 5.8, an additional reaction occurred between THSG and H 2O. Strong acids precipitated the hydrolyzation of THSG into glucose and aglycone, which then degraded into phenol compound. When strong bases existed, THSG was easily oxidized into quinine compound. Whereas it was stable in a neutral or weak basic condition, the content of THSG had not changed in 30 d. So was the THSG's crystal powder stored at room temperature for 240 d. The value of t 0.9 was 1 098 d. Conclusion THSG is stable in some certain conditions.
