摘 要:目的:研究耐力性运动对高脂饮食小鼠血浆一氧化氮以及氧化型低密度脂蛋白水平的影响,探索耐力性运动是否有可能作为药物以外的预防动脉硬化的手段。方法:实验在天津体育学院实验中心完成。采用南京建成生物工程公司试剂盒测定一氧化氮浓度,依试剂盒说明书操作。氧化型低密度脂蛋白的测定方法如下:将血浆0.3mL注入预先加入EDTA(100mmol/L)的试管中,加入柠檬酸钠和肝素的混合溶液3mL(柠檬酸钠0.064mol/L,肝素5万U/L),震荡混匀后室温放置15min,4000r/min离心15min,弃上清,在沉淀物中加入Tris-HCL0.3mL(0.02mol/L,pH值8.6)再次溶解,取再次溶解液0.3mL依次加入乙酸1.5mL(200g/L),硫代巴比妥酸1.5mL(8g/L),震荡混匀后,沸水浴60min,流水冷却后,加入正丁醇4mL,强力震荡混匀,3000r/min离心5min,532nm波长下测定吸光度。结果:高脂饮食组动物血浆一氧化氮水平犤(24.90±9.06)μmol/L犦明显低于正常饮食组犤(45.03±10.23)μmol/L犦(P<0.05),而高脂饮食结合运动干预组动物血浆一氧化氮水平犤(38.11±12.71)μmol/L犦明显高于高脂饮食组(P<0.05),并与正常饮食对照组相比无显著性差异(P>0.05)。另外,高脂饮食组动物血浆氧化型低密度脂蛋白水平犤(4.13±0.66)μmol/L犦明显高于正常饮食对照组犤(2.04±0.52)AIM:To study the effects of endurance exercise on the nitric oxide(NO) and oxidized low density lipoprotein(OX-LDL) of mice fed with high-fat food and probe into the possible mechanism in order to see whether endurance sport can be used as a way besides medicine to prevent arteriosclerosis(AS).METHODS:The experiment was completed in the Laboratory Center,Tianjin Institute of Physical Education from January to December 2002.The reagent kit of Nanjing Jiancheng Bio-engineering Corporation was used to determine the concentration of NO.The concentration of OX-LDL was determined as follows:0.3 mL plasma was added into the tube with 100 mmol/L EDTA,then 3 mL sodium citrate(0.064 mol/L) and heparin(50 000 U/L) mixture,and the mixture was stirred and placed in the room temperature for 15 minutes,then centrifuged at 4 000 rounds per minutes for 15 minutes,the suspension was discarded and 0.3 mL Tris-Hcl(0.02 mmol/L,pH 8.6) added to the setting to resolve again,then 0.3 mL solution was taken,1.5 mL acetic acid(200 g/L) and 1.5 mL TBA(8 g/L) added in turn,and the solution stirred and placed in the boiling water bath for 60 minutes,cooled down by water and 4 mL propylethanol added,stirred strongly,centrifuged at 3 000 rounds per minutes for 5 minutes,and then the absorbance(A) at 532 nm wave length was determined.RESULTS:The NO concentration of high fat diet group was(24.90 ±9.06) μmol/L,significantly lower than that of the normal diet group[(45.03±10.23)μmol/L](P< 0.05),and the NO concentration of high fat diet with exercise intervention group[(38.11±12.71)μmol/L] was significantly higher than that of high fat diet group(P< 0.05) and had no significant difference from that of the normal diet group(P >0.05).In addition,the OX-LDL concentration of high fat diet group[(4.13±0.66)μmol/L] was higher than that of the normal diet group[(2.04±0.52)μmol/L](P< 0.05),while the OX-LDL concentration of high fat diet with exercise intervention group(P< 0.05),while the OX-LDL concentration of high fat diet with exercise interven
