红参对糖尿病大鼠视网膜微血管病变细胞外基质的影响

摘　　要：目的探讨红参对糖尿病大鼠视网膜微血管病变细胞外基质的影响。方法用链脲佐菌素(65mg/kg)一次性腹腔注射的方法建立糖尿病大鼠模型,30只SD大鼠随机分成对照组、模型组和红参干预组。红参粉末(每日15g/kg)灌胃给药。210天后处死动物摘除眼球,进行血管铺片形态学观察及纤连蛋白(FN)和层粘连蛋白(LN)在视网膜血管的表达,用图像分析仪测定免疫组化显色强度,进行定量分析。结果7个月糖尿病大鼠视网膜毛细血管细胞外基质增生明显,FN和LN均呈高表达(P<005)。红参干预后FN表达比模型组明显降低(P<005),LN表达比模型组降低,但组间对比差异不显著。结论红参可抑制细胞外基质成分过度表达,从而减轻毛细血管基底膜的损伤。OBJECTIVE To study effect of red ginseng on extracellular matrix in rats with streptozotocin-induced diabetic retinopathy METHODS The model of SD rats with diabetes mellitus was set up by a single intraperitoneal injection of streptozotocin(65mg/kg) Thirty rats were randomly divided into normalcontrol group,model group and treated group with red ginseng The rats were fed with Melted red ginseng powder(1 5g/kg d-1),and were fed normal saline into stomachs in normal control group and model group All rats were killed at 210 days Retinal digest preparations were observed The expression of fibronectin(FN)and laminin(LN)were observed Quantitative analysis was performed by immunological histochemistry RESULTS At seven months proliferation of extracellular matrix was displayed and high expression of fibronectin and Laminin in the rats Compared with the model group,the expression of FN in red ginseng group was decreased evidently( P <0 05),but there was not a significant difference in LN Seven months later,the model group rats displayed apoptis of extracellular matrix and high expression of fibronectin and Laminin Compared with the model group,the expression of FN in Ginseng group was decreased evidently( P <0 05),but there was no significant diffrence of LN CONCLUSION Red ginseng could reduced damage of basement membrane and this effect may be achieved when super expression of extracellular he matrix(FN、LN)was restrained
