摘  要:目的 :分离纯化河套大黄药材活性成分Rhapontin ,建立测定Rhapontin含量的高效液相色谱法。 方法 :采用亲脂性吸附树脂与制备型RP HPLC组合分离技术 ,将河套大黄药材乙醇粗提物经亲脂性吸附树脂柱预分离 ,然后由制备型RP HPLC纯化精制。Rhapontin的测定以KromasilODS(2 5 0× 4 .6mm ,5 μm)为固定相 ,甲醇 水 (6 0∶4 0 )为流动相 ,紫外检测波长为 32 0nm。结果 :产品经熔点、EI MS、IR、UV、1 HNMR、1 3 CNMR和HPLC图谱鉴定 ,并与文献、标准对照品比较 ,证明为Rhapontin ,纯度 >98%。RP HPLC检测的线性范围为 0 .4 0 8~ 4 .0 8μg(r =0 .9999)。 结论 :RP HPLC检测方法可用于从河套大黄药材中制备Rhapontin的质量控制 ,所建立的制备力法简单 ,重复性好 ,采用的溶剂系统价廉易得。AIM: To isolate and purify the active substance Rhapontin from Rhapontineum hotaoense C. Y. Cheng et C. T. Kao and develop a HPLC for quantifying Rhapontin. METHODS: Combined lipophilic adsorption resin and preparative RP HPLC, the ethanol extractof R. hotaoense C. Y. Cheng et C. T. Kao was pre isolated by lipophilic adsorption resin, and the enriched extract was purified to give 3,3′, 5 trihydroxy 4′ methyloxystilbene 3 O β D glucoside by preparative RP HPLC. RESULTS: By contrasting with the literature data and standard substance, The 3,3′,5 trihydroxy 4′ methyloxystilbene 3 O β D glucoside was identified by El MS, IR, UV, 1HNMR, 13 CNMR, HPLC and melting point. The final purity of this product is >98%. CONCLUSION: The developed RP HPLC is simple, reproducible and easy to operate, and can be used for the quality control of preparing Rhapontin from R. hotaoense C. Y. Cheng et C. T. Kao, and the solvent is inexpensive.
