摘 要:目的 构建实用型低密度石英谐振式生物传感器 (quartz oscillation biosensor,QOBS)阵列检测系统。方法 首先利用精细微加工法制作单个 QOBS,在此基础上先后构建了粘接式以及夹具式 2× 5型 QOBS阵列 ;并自行开发研制自激式振荡电路、PESA V2 .0版频率记录分析软件 ;将它们与计算机联用以构建 QOBS阵列检测系统。结果 成功地制造出了在气、液相中稳定振荡的单个 QOBS,构建的粘接式及夹具式 QOBS阵列各有其优缺点 ;自激式振荡电路有非常强的振荡能力 ,在液相中起振非常容易 ,其频率稳定度也可达实用要求 ;PESAV2 .0版频率记录分析软件具有自行设置各初始参数 ,实时记录频率变化、自动绘制频率变化趋势图及分析结果等多项功能。结论 成功地构建出 QOBS阵列检测系统 ,搭建起一个生物检测的技术平台 ,为临床上病原性微生物的检测以及多种疾病的早期诊断提供一种新的方法。OBJECTIVETo construct detection system of practical low-density quartz oscillation biosensor (QOBS) array. METHODSTechnology of fine processing was firstly used to make the solo QOBS. Based on it, glued and clamped 2×5 style QOBS array was constructed successively. Also we developed the self-oscillating circuit and the PESA version 2.0 software applying to record and analyze frequency. So the detection system of QOBS array was constructed by combining all these components. RESULTSSolo QOBS that realized stable oscillation both in air and liquid phase was successfully constructed. Both glued and clamped style QOBS arrays had its advantages and disadvantages. And self-oscillating circuit that was developed by ourselves had strong vibromotive ability in liquid phase and its stability of frequency could also meet the request of experiment. Self-developed PESA version 2.0 software had many functions such as setting up primary parameters, recording change in frequency in real-time, plotting figure of frequency change automatically, and so on. Data would be transferred to computer automatically after the collection and analysis. CONCLUSIONSQOBS array detection system was successfully constructed. A technical platform was built for bio-detection and hence could provide a new method for the detection of pathogenic microorganism and early diagnosis for many diseases.
