摘  要:目的 :研究普鲁泊福对大鼠急性脊髓损伤的治疗作用及其机制。方法 :SD大鼠 6 0只 ,随机分为假手术组、损伤组和普鲁泊福组 (n=2 0 )。采用改良的 Allen's打击法致伤大鼠脊髓 ,假手术组仅做椎板切除术。普鲁泊福组在脊髓损伤后 30 min以 6 0 mg/ (kg· h)的速度持续输注 1%的普鲁泊福 1h,其余两组以相同的方法给予 0 .9%的生理盐水。应用斜板试验和 BBB(Basso,Beattie and Bresnahan)评分法进行后肢运动功能评价 ,分别采用黄嘌呤氧化酶法和硫代巴比妥酸法测定血浆超氧化物歧化酶 (SOD)活力和丙二醛 (MDA)含量变化 ,同时观察组织形态学变化。 结果 :普鲁泊福组行为学评分明显高于损伤组(P<0 .0 5 ) ;与假手术组相比 ,损伤组血浆 MDA含量升高 ,SOD活力降低 ,而与损伤组相比 ,普鲁泊福组血浆 MDA含量降低 ,SOD活力升高 ;损伤组脊髓病理变化较重 ,可见大量神经元坏死 ,普鲁泊福组神经元坏死较轻。 结论 :普鲁泊福对大鼠急性脊髓损伤有较好的治疗作用 ,其机制与抗氧化作用有关。Objective:To study the neuroprotective effect and its mechanism of propofol on acute spinal cord injury in rats. Methods:Sixty Sprague-Dawleys rats were randomly divided into 3 groups:sham operation group, injury group and propofol treatment group. In propofol group, 1% propofol was infused at a rate of 60 mg/(kg·h) for an hour, while 0.9% saline was used in other groups. Rat models of spinal cord contusion were made by a modified Allen's method. The inclined plane and BBB(Basso,Beattie and Bresnahan) scales were used to assess hind limb neurological function. Superoxide dismutase(SOD) activity and malonaldehyde(MDA) level in the plasma were measured by thiobarbituric acid(TBA) and xanthine oxidase methods respectively. Pathological changes were observed at the same time. Results: Behavioral score of propofol group was significantly higher than that of injury group. Compared with sham operation group, MDA level in injury group was increased and SOD activity was reduced. Compared with the injury group, MDA level in the propofol group was much lower and SOD activity increased remarkably. In injury group, a large number of neuronal death could be seen and pathological changes in propofol group were better than those in injury group. Conclusion:Propofol has neuroprotective effect on acute spinal cord injury, which is possibly related to its antioxidation function.
