摘  要:目的 探讨血管抑素蛋白对体外培养的鼠视网膜微血管内皮细胞 (REC)生长的抑制作用及其抑制视网膜新生血管形成的效果。方法 从血浆中分离血管抑素。原代培养鼠视网膜微血管内皮细胞 ,四甲基偶氮唑盐 (MTT)比色法检测不同浓度的血管抑素对其生长的抑制作用。高浓度氧诱导C57BL/ 6J小鼠建立视网膜缺血性病变动物模型。将 30只小鼠分为 5组 :正常组 ;高氧对照组 ;实验Ⅰ组 (血管抑素浓度 15 0 μg/ μl) ;实验Ⅱ组 (血管抑素浓度 2 0 0 μg/ μl) ;实验Ⅲ组 (血管抑素浓度 30 0μg/ μl) ,计数各组小鼠视网膜新生血管内皮细胞核数 ,并加以比较。 结果 血管抑素在体外能显著抑制视网膜微血管内皮细胞增殖 ,其半数有效剂量 (ED50 )约为 130 μg/ml。吸入高氧的鼠不论是对照组还是血管抑素治疗组每只眼均可见突出内界膜的新生血管内皮细胞 ,发生率为 10 0 %。实验Ⅰ、Ⅱ、Ⅲ组与高氧对照组相比小鼠视网膜新生血管细胞核数分别减少了 4 2 %、5 7%、82 %。结论 血管抑素在体外能有效地抑制视网膜微血管内皮细胞增殖。Objective To observe the inhibitory effects of angiostatin on microvascular endothelial cells of mouse retina and test the efficacy of native angiostatin in suppressing experimental retinal neovascularization induced by oxygen Methods Angiostatin was purified with L lysine Sepharose 4B from human plasma The primary microvascular endothelial cells from rat retina were cultured Microvascular endothelial cells growth inhibition assay was carried out with MTT method Mouse models of hyperoxia induced ischemic retinopathy were established Angiostatin or normal saline (NS) were injected into the vitreous in 5 groups: normal, control and various doses The nuclei of new vessel buds extending from the retina into the vitreous in different groups were counted and compared under the light microscope Results Angiostatin could inhibit the growth of microvascular endothelial cells from rat retina in vitro There were plenty of new vessel buds in the eyes of all mice in hyperoxic condition The number of the nuclei of new vesselbuds in the murine eyes with injection of angiostatin They were reduced by 42%( P <0 01), 57% ( P < 0 01) and 82%( P <0 01) respectively Conclusion Angiostatin can powerfully inhibit growth of microvascular endothelial cells The proliferation of retinal vessel may be suppressed by using angiostatin
