FK506抑制心脏移植急性排斥反应对[Ca^(2+)]i影响的实验研究

摘　　要：目的　探讨心脏移植急性排斥反应心肌细胞内 [Ca2 + ]i的表达及FK5 0 6在发挥免疫抑制作用时对其的影响。方法　利用大鼠异位心脏移植建立急性排斥反应模型 ,借助流式细胞仪 ,用钙离子敏感性荧光探针 (Fluo 3 )检测移植心脏心肌细胞内 [Ca2 + ]i荧光强度 ,检测 [Ca2 + ]i的变化规律及FK5 0 6对其的影响。结果　同种异体移植组心肌细胞内 [Ca2 + ]i荧光强度 ( 12 1.96± 3 4.93 )明显高于同系异体移植组 ( 3 4.11± 19.2 8,P <0 .0 1)。FK5 0 6能够抑制急性排斥反应心肌细胞内 [Ca2 + ]i的升高 ( 15 .97± 7.2 5 ,P <0 .0 1)。结论　心脏移植急性排斥反应中心肌细胞内 [Ca2 + ]i明显升高。FK5 0 6能够防止 [Ca2 + ]i升高 。Objective To investigate the expression of intracellular calcium in acute cardiac allograft rejection and the effect of FK506 on it.Methods Fluorescent intensity of intracellular calcium in cardiac myocytes was measured by flow cytometry with a calcium sensitive fluorescent probe (Fluo-3) in heterotopic rat heart allografts (Dahlto SD) and isografts (SD to SD) with or without chronic immunosuppression by FK506.Results On the 5th day, fluorescent intensity of intracellular calcium in cardiac myocytes was significantly increased in allografts (P<0.01),as compared with isograft hearts (121.96±34.93 versus 34.11±19.28,respectively) and chronic administration of FK506 (0.15 mg·kg -1·d -1 28 days) to animals with cardiac allografts suppressed the increase of intracellular calcium (15.97±7.25(P<0.01).Conclusion The intracellular calcium in cardiac myocytes obviously increases during acute allograft rejection. FK506 can prevent the increase and suppress the intensity of acute rejection in cardiac allografts.
