反复自然流产患者血清及绒毛组织细胞因子研究

摘　　要：目的　对比观察反复自然流产组 (RSA)与正常妊娠人工流产组患者血清中一氧化氮 (NO)含量、一氧化氮合酶 (NOS)活性和血管内皮生长因子 (VEGF)含量及绒毛组织中NOS活性、P5 3蛋白及bcl 2蛋白的表达水平 ,为RSA的防治提供依据。方法　 3 3例RSA患者及 3 0例正常妊娠人工流产者血清NO含量、血清及绒毛组织中NOS活性检测 ,均采用比色法进行 ;血清VEGF含量检测采用ELISA方法 ;绒毛组织中P5 3蛋白及bcl 2蛋白的表达水平采用流式细胞仪检测。结果　RSA组血清NO含量较正常妊娠人工流产组升高 ( P <0 .0 5 ) ;血清NOS活性较对照组升高 ( P <0 .0 1) ;绒毛组织中NOS活性较对照组升高 (P <0 .0 1) ;血清VEGF含量较对照组升高 (P <0 .0 1) ;绒毛组织中P5 3蛋白表达 ,较对照组升高 (P <0 .0 1) ;绒毛组织中bcl 2蛋白表达 ,较对照组降低 (P <0 .0 1)。结论　RSA组患者血清中NO含量、NOS活性及VEGF含量和绒毛组织中NOS活性、P5 3蛋白表达量均明显升高 ,bcl 2蛋白表达量明显减低 ,均可导致胎盘细胞凋亡增强 。Objective To investigate the content of NO,VEGF, activity of NOS in serum,the expression level of P53 and bcl-2 and activity of NOS in chorionic villi in patients with recurrent spontaneous abortion (RSA) and normal abortion,in order to provide basis for prevention and treatment of RSA.Methods 33 patients with RSA and 30 women with normal abortion were divided into RSA group and control group.The activity of NOS in chorionic villi was detected by eolorimetry.VEGF content in serum was determined by ELISE.The P53 and bcl-2 expressions were detected by flow cytometry and immunofluorescent technique.Results As compared with those in control group the content of serum NO in RSA group was obviously increased (P<0.05);NOS activity in serum was significantly increased (P<0.01);the NOS activity in chorionic villi was markedly increased(P<0.01);the VEGF content in serum was significantly increased (P<0.01),The expression level of P53 protein in chorionic villi was notably increased (P<0.01);the expression level of bcl-2 protein in chorionic villi was significantly decreased (P<0.01).Conclusion The changes of cytokines in serum and chorionic villi in patients with RSA can result in the increase of apoptosis of placental cells,which may be related to the pathogenesis of RSA.
