ISCOMs作为粘膜免疫佐剂在避孕疫苗构建中的作用研究

摘　　要：目的　探索有效提高避孕疫苗生殖道粘膜免疫的方法。方法　将合成纯化的 3 5多肽与皂甙QuilA、脂类混合物 (等体积比的磷脂和胆固醇 )以 1∶4∶1比例混合 ,经充分透析后得到 3 5肽 ISCOMs ;将 3 5肽 ISCOMs和 3 5肽 弗氏佐剂混悬物分别免疫纯系BALB c雌性小鼠 ,并比较免疫后小鼠生殖道内特异性IgA的动态变化以及小鼠生育力的变化。结果　制备成功的 3 5肽 ISCOMs在电镜下可见明显的、较规则的蜂巢样结构 ;3 5肽 ISCOMs免疫组IgA抗体在第 10周时达最高值 ( 1∶60 0 ) ,约持续 4周左右 ;而 3 5肽 弗氏佐剂组IgA抗体在第 14周时达最高值 ( 1∶3 0 0 ) ,约持续 1周左右 ;3 5肽 ISCOMs免疫后小鼠的受孕率 ( 14 3 % )显著低于 3 5肽 弗氏佐剂免疫组 ( 60 0 % )。结论　ISCOMs可以显著提高抗原在生殖道内的粘膜免疫效应 ,增强抗生育作用 ,是一种极富潜力的粘膜免疫佐剂。Objective To explore the methods for the improvement of the effect of genital mucosal immunologic reaction of the contraceptive vaccine. Methods A chimeric peptide with 35 amino acid residues and two specific epitopes was obtained by chemical synthesis. After purification, the peptide was mixed with Quil A and lipid mixtures at the ratio of 1∶4∶1 to form immune-stimulating complexes (ISCOMs) after sufficient dialysis. C57B1/6 mice were divided two groups A and B, in which mice were immunized respectively by peptide-ISCOMs and peptide-Freund's adjuvant. Results Regular and honeycomb-like structures of peptide-ISCOMs were found by electron microscopy. The highest titer of specific IgA in the vaginal lotion lasted for about 4 weeks in group A at 1∶600, but just 1 week in group B at 1∶300. The rate of pregnancy in group A (14.3%) was significantly lower than that in group B (60.0%). Conclusion ISCOMs could significantly improve the effect of genital mucosal immunologic reaction.
