摘  要:目的 探讨中药复方—— 地甘口服液对辐射损伤小鼠骨髓细胞周期素基因及造血干细胞和基质细胞粘附分子CD45、VCAM-1表达的作用。方法 建立辐射损伤模型(一次性用直线加速器给予7.0 GY的X线照射)。喂饲100%地甘口服液每次0.2 mL/10g小鼠,每日2次。第10天用原位杂交法检测小鼠骨髓单个核细胞cyclin D1 mRNA的表达,用免疫组化SABC法检测骨髓VCAM-1的表达,用流式细胞仪检测CD45的含量。结果 地甘口服液组表达明显高于模型组,前两者与正常组无明显差异,CD45的表达仍未达到正常水平。结论 中药复方——地甘口服液能增强辐射损伤小鼠骨髓细胞周期素基因和造血细胞及基质粘附分子(CD45、VCAM-1)的表达,促进造血细胞的增生。Objective To study the effect of compound TCM on expression of cylin D1 mRNA and adherent molecule VCAM-1 in bone marrow cells and stromal cells in radiation lesion mice. Methods After the model radiation lesion given 7.0 Gy of X ray by linear accelerator was established, the animals were fed with 100% Digan Oral Liquid 0.2 mL/10 g mouse, twice a day for 9 days. At tenth day cyclin D1 mRNA expression level of marrow MNC in model mice was measured by in situ hybridization, and VCAM-1 was detected in sromal cells by immunohistochemistry SABC, and expression of CD45 in bone marrow MNC was measured by flow cytometry. Results Expression levels of cyclin D1 mRNA, VCAM-1 and CD45 in DGOL group were significantly higher than that in the model group, and both of before them were similar to that in the normal group, expression of CD45 was lower than that in the normal group. Conclusion Compound TCM DGOL can increase the expression of cyclin D1 mRNA and adherent molecule VCAM-1 in bone marrow radiation lesion and promote the growth of hematopoietic cells.
