摘　　要：目的 :建立同时测定血浆中地西泮及其代谢物浓度的方法。方法 :选用ZORBAXRP C18柱 (15 0mm× 4 6mm ,5 μm) ;甲醇 - 2 5mmol·L-1醋酸铵溶液 (6 0∶4 0 ,V/V)作流动相 ;流速 0 8mL·min-1;检测波长 2 30nm。取血浆样品 0 5mL ,在碱性条件下用二氯甲烷 -正己烷提取 ,HPLC检测。结果 :本法对替马西泮、去甲地西泮和地西泮 3种物质的最低检测限均为 2 μg·L-1,线性范围为 10～ 15 0 0 μg·L-1;奥沙西泮的最低检测限为 5 μg·L-1,线性范围为 2 0～ 15 0 0 μg·L-1。回收率均接近 10 0 % ,日内、日间RSD <5 %。结论 :本法能同时测定血浆中地西泮及其代谢物浓度 ,具有重现性好 ,灵敏、可靠 。AIM: To establish a method to determine simultaneously diazepam and its metabolites in plasma by HPLC. METHODS: The ZORBAX reversed-phase C 18 colume was used,the mobile phase consisted of methanol-25 mmol·L -1 NH 4Ac(60∶40,V/V),the flow rate was 0.8 mL·min -1,the absorbance of eluate was monitored at 230 nm.Alkalized plasma sample was extracted with n-hexanedichloromethane,and injected into HPLC system. RESULTS: The detection limit of this method was 5 μg·L -1 for oxazepam,2 μg·L -1 for temazepam,nordiazepam and diazepam,the method recovery approached 100%,RSD was less than 5%,and their linearity range was 20-1 500 μg·L -1 in plasma. CONCLUSION:This sensitive and selective method can be used to determine diazepam and its metabolites in plasma.This method has good recovery and reliability.It can be helpful in therapeutic drug monitoring and in the control of abuse drugs.
