载脂蛋白H单克隆抗体的制备与鉴定

摘　　要：目的制备及鉴定载脂蛋白HApoH单克隆抗体。方法从人血清中分离纯化ApoH免疫小鼠后取其脾细胞与骨髓瘤细胞融合,经筛选出两株杂交瘤细胞株2C5和3H4,制备腹水后,用硫酸胺沉淀等方法纯化,获得ApoH单抗。结果两株细胞产生的单抗皆属于IgG1,与其它载脂蛋白无交叉反应,细胞株连续传代3个月仍能稳定分泌ApoH单抗,且效价稳定。结论2C5和3H4两株细胞能分泌合格的ApoH单抗。objective:Preparation of ApoH McAb.Methods:Separation and purification of ApoH from human serum,After immunization of rats using ApoH,its splen cells and myeloma were fused.Two hybridoma cell lines(2C 5 and3H 4 )were selected.After prepration of ascrites the aseritf was purificat using ammonium sulfate precipition(NH 4 )2SO 4 methods and than ApoH McAb was obtained.Rusults:The McAb of both cell lines belong in IgG 1 and had not cross reaction with other apoproteins.These cell lines still could stably secrete ApoH McAb and the effect value was stabile after three months.Conclusions:2C 5 and3H 4 cell lines can secrete qualified ApoH McAb.
