摘 要:目的:观察水杨酸钠腹腔注射制备的耳鸣模型动物下丘外侧核单单位自发放电变化和应用尼莫地平对其影响。方法:应用水杨酸钠(400mg/kg)腹腔注射制备豚鼠耳鸣模型,采用活体细胞外单位记录法观察了耳鸣模型组、对照组和尼莫地平组豚鼠用药前后下丘外侧核229个神经元单单位自发放电率变化,并采集耳鸣模型组神经元特征放电模式进行分析。结果:耳鸣模型组在水杨酸钠注射后2h下丘外侧核单单位自发放电率为(17.6976±1.2226)Hz,较注射前(9.4116±1.0573)Hz明显增加,差异有极显著性意义(F=6.216,P<0.01),并且出现连续4个波峰以上的簇状波。对照组等量生理盐水注射前后下丘外侧核单单位自发放电率差异无显著性意义(P>0.05),也未见连续4个波峰以上的簇状波。对照组与耳鸣模型组相比差异有极显著性意义(F=5.704,P<0.01)。尼莫地平注射后下丘外侧核单单位自发放电率为(7.9883±1.1832)Hz,与耳鸣模型组水杨酸钠注射后相比差异有极显著性意义(F=6.437,P<0.01)。尼莫地平注射能抑制水杨酸所致下丘外侧核单单位自发放电率的增加和连续4个波峰以上的簇状波的出现。结论:水杨酸所致的豚鼠下丘外侧核单单位自发放电的变化,尤其是连续4个峰以上的长簇状波可能是一种耳鸣相关的神经现象。尼莫地平对水杨酸所致的豚鼠下丘外侧核?AIM:To observe the spontaneous discharge of single-unit neurons in the inferi or colliculus(IC) of an animal model of tinnitus established by intraperitoneal injection of sodium salicylate in order to elucidate the electrophysiologic mech anism of tinnitus,and furthermore to evaluate the effect of nimodipine. METHODS:The spontaneous activities of 229 units from lateral nuclei in the IC were recorded with extracellular recording in 24 guinea pigs,which were randomly divided into control group,tinnitus model group and nimodipine group.The charac teristics of the abnormal pattern of discharges were analyzed in the tinnitus mo del group. RESULTS: Two hours after salicylate injection,the spontaneous activity was sig nificantly increased from(9.411 6±1.057 3) Hz before treatment to(17.697 6±1.2 22 6) Hz in the model group, with significant differences(F=6.216,P< 0.01).And b urst discharge with more than 4 continuous spikes was found in the tinnitus mode l group. But comparison of spontaneous activity recorded before and after saline administration in the control group revealed no statistically significant diffe rences(P >0.05),and no burst discharge with more than 4 continuous spikes was fo und. Spontaneous activity recorded after nimodipine administration were (7.988 3 ±1.183 2) Hz,which revealed highly significant difference from that in the tinn itus model group after salicylate administration(F=6.437,P< 0.01).Nimodipine inh ibited the increase of the spontaneous activity and presence of burst discharge with over 4 spikes induced by salicylate. CONCLUSION:The changes of spontaneous activity of single unit of external nucl eus in IC due to salicylate administration,especially burst discharge with more than 4 continuous spikes, may be associated with tinitus-related neuronal activ ity.Nimodipine induced inhibition for the spontaneous activity may be related wi th the therapeutic action of nimodipine on tinnitus.
