摘 要:目的 :观察中药肝力克对肝纤维化大鼠肝组织iNOSmRNA (诱生型一氧化氮合酶mRNA)表达的影响 ,探讨其作用机制。方法 :用 40 %CCl4皮下注射建立大鼠肝纤维化模型。造模结束后 ,给予不同剂量的肝力克和秋水仙碱灌胃治疗 9周。采用原位杂交法观察大鼠肝组织中iNOSmRNA的表达情况。结果 :肝纤维化模型组大鼠肝组织iNOSmRNA阳性表达较正常对照组明显升高 (P <0 0 1) ;应用肝力克能明显改善大鼠肝组织炎症活动度和纤维程度 ,能明显下调肝纤维化大鼠肝组织iNOSmRNA表达。结论Objective:To investigate the mRNA expression of the inducible nitric oxide synthase and the effects of Ganlike in rats with experimental liver fibrosis induced by carbon tetrachloride. Methods:SD rats′model of fibrosis was induced by injecting CCl 4. After model being built, they were protected with Ganlike and Colchicine, and normal saline were taken as the control. The expression of iNOS mRNA in all liver tissues was detected by in situ hybridization method. The severity of inflammation and fibrosis in all liver tissuses were measured by semiquantitative scoring. Results: the expression of iNOS mRNA in the hepatic fibrosis were significantly higher than the control group(P<0.01), after the treatment with Ganlike, The severity of inflammation and fibrosis were obviously improved, and the expression of iNOS mRNA in hepatic fibrosis was downregulated. Conclusion:Ganlike can downregulate the expression of iNOS mRNA in hepatic fibrosis, which maybe a possible mechanism of its anti-hepatic fibrosis.
