摘  要:目的观察罗格列酮对单侧输尿管梗阻(UUO)大鼠肾皮质过氧化物酶体增殖物活化受体(PPAR)γ、转化生长因子(TGF)β1表达介导的肾间质纤维化的作用。方法UUO大鼠给予罗格列酮5mg·kg-1·d-1灌胃,用免疫组化、RT-PCR及Western印迹的方法检测术后7d、14dPPARγ、TGF-β1、增殖细胞核抗原(PCNA)表达量及观察肾脏病理改变。结果与假手术组相比,UUO组及药物治疗组PPARγ、TGF-β1、、PCNA表达均增高且UUO组显著高于治疗组(P<0.05)。结论罗格列酮可通过活化PPARγ,下调TGF-β1,从而减轻UUO术后肾组织间质纤维化。Objective To investigate the effect of rosiglitazone on the expr es sion of PPAR?,TGF-a1 and PCNA and its mechanism on renal interstitial fibrosi s following unilateral ureteral obstruction(UUO) in rat kidney. Methods Thirty r ats were randomly assigned to shame operation group(sham group),UUO group, rosig litazone(5 mg穔g-1·d-1) treatment group after UUO. Immunohistochemistry,RT-PCR,Western blotting were performed to investigate renal pathological changes an d examine the expression of PPAR?,TGF-a1,PCNA on the 7th and 14th day in the kidney. Results In comparison with the shame group,the expression of PPAR?,TGF-a1,PCNA of UUO and treatment groups increased significantly(P< 0.05),meanwhil e rosiglitazone could significantly decrease the expression of PPAR?,TGF-a1,P CNA as compared to UUO group(P< 0.05). Conclusion Rosiglitazone may play a prote ctive role of the kidney by activating PPAR?,downregulating TGF-a1 and allevi ating renal interstitial fibrosis following UUO.
