疲劳小鼠骨骼肌中差异表达基因的筛选和鉴定

摘　　要：选用 30只普通级 BAL B/ c雄性小鼠为试验对象 ,随机分成对照组、浸水组和游泳疲劳组。利用改进后的 m RNA差异显示法筛选疲劳小鼠骨骼肌中的差异表达基因 (DD- ESTs) ,经反向 Northern blot和测序鉴定后 ,获得了 4条阳性目的片段 ,其中有 3条 DD- ESTs只在游泳疲劳组表达 ,另一条呈现下调表达现象 ,登录 Gen Bank数据库获得登录号为 AY5 4 36 4 9、AY5 4 36 5 0、AY5 4 36 5 1和 AY5 4 36 5 2 。BALB/c mice were selected as experimental object and divided into negative control group, positive control group (dipping in the water) and swimming-fatigue group respectively. The differentially expressed genes in mice skeletal muscles were screened with improved mRNA differential display method, and were evaluated by Reverse Northern blot and sequencing. Four segments expressed obviously differentially were obtained and three of them expressed only in swimming-fatigue group and the other one expressed by down-regulation. Through the GenBank date base to Register, accession numbers were obtained, which were AY543649,AY543650,AY543651,AY543652. The exact relationship between the four segments and fatigue remains to be investigated further.
