猪原代肝细胞的分离及体外培养模型的建立

摘　　要：采用剪切消化法 ,对猪肝细胞进行了分离和原代培养。结果显示 :分离的肝细胞即时存活率为 93.4 % ,培养存活率为 87.6 % ,并观测了肝细胞体外的生长形态和增殖规律 ,摸索了建立猪原代肝细胞体外培养模型的条件 。Porcine hepatocytes were isolated and cultured primarily according to Luben′s technique with slight modifications.The results showed that the immediate survival rate of these cells separated was 93.4% and the cultured survival rate reached 87.6%.Based on the morphological alteration and the proliferation regularity of hepatocytes in a culture,the conditions of culturing in vitro were explored successfully.Therefore,this study may provide an important information for researching the porcine nutrient metabolism on the basis of cell levels.
