摘  要:目的 研究肝细胞生长因子及其受体在骨髓干细胞移植治疗中的表达情况 ,评价其作用。方法 大鼠 30只 ,其中 15只接受了冠状动脉前降支结扎以引起前壁心肌梗死 ,另外 15只同时还进行了骨髓单个核细胞心肌移植。术后不同时间点心脏取材检测肝细胞生长因子及其受体的表达。结果 心肌梗死之后心肌组织肝细胞生长因子及其受体表达水平增加 ;同时接受骨髓单个核细胞心肌移植治疗没有进一步增加肝细胞生长因子的表达 ,但其受体的表达水平进一步显著增加 (P <0 0 1)。结论 骨髓单个核细胞心肌移植治疗能够增加肝细胞生长因子受体的表达水平。Objective Aim to investigate the role of hepatocyte growth factor(HGF) in the course of bone marrow mononuclear cells transplantation. Methods Thirty male Lewis rats were accepted to perform operation, 15 were only performed liga tion in the left anterior descending coronary artery (LAD), the other 15 were al so performed bone marrow mononuclear cells transplantation after the ligation of LAD. At 1 day, 3 days, 1 week, 2 weeks and 4 weeks after the operation, the hea rts were taken out for the detection of HGF and c-met by using PCR method. Norm al rat′s heart was also tested for HGF and c-met. Results _Th e normal heart had HGF expression, and after ligation of coronary artery, the HGF expression level of the heart increased. With bone marrow mononuclear cells transplantation the HGF expression level did not change further. Normal heart ha d little c-met expression. After AMI, the level of c-met expression increased. With bone marrow mononuclear cells transplantation, the increase of c-met expr ession was higher than that with AMI only (P<0.01). Conclusions Transplantation of bone marrow mononuclear cells into the heart couldn′t increase the HGF expression further than that after AMI only, but could increa se the c-met expression in the heart.
