摘  要:通过RT PCR方法,分别从两栖类的黑斑蛙(Rananigromaculata)和鸟类的鹌鹑(Coturnixcoturnixjaponica)中首次克隆了核糖体大亚基蛋白L15(RPL15,ribosomalproteinL15)的cDNA片段,并分析了其序列特征.黑斑蛙和鹌鹑的rpl15cDNA片段分别为467个核苷酸和408个核苷酸,推测分别编码155个氨基酸和136个氨基酸的蛋白片段,与其它已知的脊椎动物rpl15序列有高度的同源性.应用这两条cDNA序列和其它已知的脊椎动物rpl15序列构建的系统发育树显示聚类结果与传统分类一致.The cDNA fragments of ribosomal protein L15 (RPL15) were cloned respectively from Rana nigromaculata and Coturnix coturnix japonica by RT-PCR. The two cDNA fragments are individually composed of 467 bp and 408 bp, encoding a protein fragment of 155 aa and another one of 136 aa. Alignment analysis indicated that both the two nucleotide sequences and the deduced amino acid sequences show high identity to other known rpl15 sequences from vertebrates. The two cDNA sequences were used to construct phylogenetic trees with other known vertebrate rpl15 sequences, and the obtained trees demonstrated similar topology with the classical systematics.
