摘 要:目的:建立高效液相色谱法(HPLC),测定甲钴胺的有关物质。方法:采用Lichrospher-NH2色谱柱(250 mm × 4.6 mm,5μm),流动相为0.3%磷酸溶液:乙腈(32:68),流速1.0 mL/min,检测波长254 nm。结果:甲钴胺、氰钴胺和羟钴胺的最 低检测限量分别为4.4、2.6和4.0 ng,甲钴胺与各杂质分离良好。结论:该法简便、快速,灵敏度高,准确可靠。Objective: To establish high performance liquid chromatography (HPLC) for the determination of the mecobalamin-related substances. Methods: The Lichrospher-NH2 chromatographic column (250 mm×4.6 mm,5μm) was used. The mobile phase was 0.3% H3PO4: acetonitrile sohition(32:68), the flow rate was 1.0 ml/ min and the detection wavelength was 254 nm. Results: The lowest detection limits for mecobalamin, hydroxobal-amin and cyanocobalamin were 4.4, 2.6 and 4.0 ng, respectively. Mecobalamin and the related substances were well isolated. Conclusions: The method is simple, rapid, highly sensitive, accurate and reliable.
