洛沙坦对兔静脉移植物内膜增生的影响

摘　　要：目的 探讨血管紧张素Ⅱ受体拮抗剂洛沙坦对静脉移植物内膜增生的影响。方法 新西兰大白兔20只,随机分为治疗组和对照组。术后治疗组即给予洛沙坦纯粉25mg/(kg·d),对照组常规喂养,共4周。术前及处死动物前分别测定基础血压和治疗后血压;术后4周测定新生内膜面积、内膜中膜比。结果 治疗组新生内膜面积(0.51±0.05)mm^2较对照组(1.02±0.10)mm^2明显减少,P<0.01。治疗前后血压无显著变化。结论洛沙坦对静脉移植物内膜增生有明显的抑制作用。Purpose: To investigate the effect of Losartan, a nonpeptide antagonist of angiotensin type 1 (AT1) receptor, on neointima formation in venous graft. Methods: Twenty New Zealand rabbits were randomly divided into 2 groups, treatment group: 25 mg &middot kg-1 &middot d-1 orally at the time of postoperation, and control group: provied with normal rabbits chow. External jugular vein of New Zealand rabbits was harvested and grafted into the ipsilateral carotid artery. Mean blood pressure was monitored before drug treatment(base pressure), and before sacrifice(post-treatment pressure). The venous grafts were harvested at the fourth week of postoperation. The cross sectional areas were calculated. Results: The neointimal areas were significantly reduced in treatment group than control group(P < 0.01). No significant difference of mean pressure was found between base pressure and post-treament pressure in treatment group (p = NS). Conclusions: Losartan couold prevent the intimal hyperplasia in venous graft and don't change normal animal's blood pressure.
