Random Amplified Polymorphic DNA Analysis of Near-isogenic Lines Differing for a Powdery Mildew Resistance Gene in Wheat

摘  要:以10个核苷酸长的随机引物,通过多聚酶链式反应(PCR)技术对1对小麦抗白粉病近等基因系的基因组DNA进行了扩增,旨在鉴定出与抗白粉病基因连锁的分子标记。对100个随机引物进行了筛选,发现有4个引物(OPV03,OPV06,OPW15和OPX05)在这对近等基因系间表现出扩增产物的多态性,其中由引物OPV03和OPV06扩增的多态DNA片段OPV03—1950和OPV06—1150只出现在抗病的近等基因系Khapli/8 chancellor中,推断其与抗白粉病基因连锁,但其连锁关系仍有待通过F2分离群体分析进一步确定。A technique based on a recent modification of the polyerase chain reaction (PCR) was used to amplify random sequences from one pair of wheat near-isogenic lines that differed for a powdery mildew resistance gene(Pm4a) in order to identify the randomly amplified polymorphic DNA(RAPD) markers linked to the gene for resistance to powdery mildew. One hundred 10-mer oligonucleotide primers were screened and four of them (OPV03, OPV06,OPW15 and OPX05) showed amplified product polymorphisms between the near-isogenic lines. Two polymorphic DNA markers (OPV03-1950 and OPV06-1150)only present in the resistant line could be linked to the Pm4a gene,but further investigation of segregating F2 population should be conducted to confirm the linkage between these DNA markers and the resistance gene.
