摘 要:从19种抗登革病毒单克隆抗体(简称单抗)中筛选出4种具有中和活性的单抗。经过纯化免疫新西兰大白兔制备多克隆抗独特型抗体(Ab2)。制备的4种抗独特型抗体血清有很高的特异性,其中具有全交叉反应的3种单抗的抗血青中Ab2的水平较高。采用简便的去除抗血清中的同种型和同种异型抗体的方法,用夹心ELISA法检测Ab2的水平取得了满意的结果。Four monoclonal antibodies (MAb) screened from 19 MAbs against dengue virushad neutralization to the virus. Three of them had cross-reactivity with other 3 dengue virus types.Individual New Zealand white rabbits were immunized with one of the four purified MAbs to gen-erate polyclonal anti-idiotypic antibodies (Ab2) to these MAbs. Four anti-idiotypic sera had highspecificity against their immunogens. Three immune sera to the cross-reactive MAbs had hightiters of Ab2. A simple procedure was used to remove allotypic and isotypic antibodies in the Ab2sera. The results, obtained by using the sandwich ELISA method to detect Ab2 Ievels in the treatedsera, were satisfactory.
