摘  要:目的研究低剂量铅对原代培养的海马神经元神经细胞黏附分子(NCAM)表达的影响。方法将孕18dWistar大鼠麻醉后剖腹取出胎鼠,建立原代培养的海马神经元,与不同剂量的氯化铅(PbCl2)共孵育后,应用免疫印迹法技术检测培养第1至7天海马神经元NCAM的表达。结果正常条件下培养的海马神经元,第1天NCAM的表达极其微弱,积分吸光度值为14,NCAM表达的高峰时间在培养第3~5天,积分吸光度值为2542~2580;随培养时间的延长,NCAM表达逐渐降低。在铅的作用下,NCAM的表达在培养早期(第2~4天)受到抑制,且呈现明显的剂量效应关系,随时间延长,抑制程度逐渐减弱;培养至第5天,铅的抑制作用消失,10-2、10-3及10-4mmol/LPbCl2剂量组NCAM的表达甚至超出了对照组;此后,所有组的NCAM表达出现下降趋势,铅对NCAM表达的抑制又表现出剂量效应关系。结论低剂量铅暴露显著抑制了原代培养的大鼠海马神经元NCAM的表达,并造成了海马神经元NCAM表达时程上的延迟。Objective To study the impact of low level lead exposure on neural cell adhesion molecule (NCAM) expression of primarily cultured hippocampal neurons Methods Wistar rats gestated at 18 th day were anaesthetized and paunched to get the pups,the hippocampi of the pups were separated and the hippocampal neurons were primarily cultured After co cultivated with different dosage of PbCl 2, the NCAM expression of the neurons were tested with Western blotting at different culture time Results Normally, the expression of NCAM at the 1 st culture day was very low and its’ integral obsorbency density was 14; the climax expression time of NCAM of the cultured hippocampal neurons was 3 rd to 5 th cultured day, and their integral obsorbency density were 2 542 to 2 580; henceforth, the NCAM expression declined NCAM expression was inhibited significantly by lead during the 2 nd to 4 th cultured day, and dose response relationship was observed The inhibition of lead weakened along with the cultured time prolonged, at 5 th cultured day,it disappeared, and the NCAM expression of 10 -2 ,10 -3 and 10 -4 mmol/L groups even exceded the control groups After that,the expression of NCAM in all groups began to decline,and the dose rosponse relationship of lead to the NCAM expression was observed again Conclusion Low level lead might significantly inhibit the NCAM expression of the primarily cultured Wistar rats’ hippocampal neurons, and might delay the climax NCAM expression time
