HPLC法同时测定黄檗皮中小檗碱、掌叶防己碱和药根碱的含量

摘　　要：建立一种高效快速从黄檗皮中同时测定小檗碱、掌叶防己碱、药根碱含量的方法。采用高效液相色谱法 ,以乙腈∶水为 1∶1(10 0 0mL溶液中含磷酸二氢钾 3.4g及十二烷基磺酸钠 1.7g)为流动相 ,流速 1.0mL/min ;检测波长为 345nm。在上述色谱条件下 ,3种生物碱达到分离完全 ,且峰形良好。该方法简单、快速、准确测定原料中 3种生物碱含量 ,为原料的质量控制奠定了基础。A method with high efficiency and rapidness for synchronously determining the contents of berberine, palmatine and jatrorrhizine from Phellodendren amurense bark was developed. With HPLC method, using KYA HIQ C 18 column as fixed phase and acetonitrile and water with the proportion of 1∶1(1 000 mL solution contains KH 2PO 4 3.4 g and CH 3 (CH 2) 11 SO 3Na 1.7 g) as mobile phase, threes alkaloids were fully separated by means of detecting at the wavelength of 345 nm with a flow rate of 1 mL/min for the eluate. This method is suitable for assaying the contents of three alkaloids in raw P. amurense bark, which will establish a basis for quality control of raw material.
