大鼠角朊细胞增殖活性的影响因素

摘　　要：目的 :探讨大鼠角朊细胞增殖活性的影响因素。方法 :将传代培养的第 2代大鼠角朊细胞 (KCs) ,以含有新生牛血清的 DMEM- F12培养基进行无滋养层法培养 ,当细胞培养至对数生长期时 ,分别改用含有不同浓度表皮生长因子 (EGF)、肝细胞生长因子 (HGF)、成纤维细胞生长因子 (FGF)、霍乱毒素 (CT)、氢化可的松(HVB)、胰岛素 (Insulin)、三碘甲腺原氨酸 (T3)的培养液继续培养 ,以 MTT法测定细胞增殖活性。结果 :在一定浓度范围内 ,EGF (5～ 2 5 μg· L- 1 )、 HGF (0 .1～ 10 μg· L- 1 )、 FGF (0 .0 5～ 2 .0 μg· L- 1 )、 CT (2～10μg· L- 1 )、 HVB (0 .1～ 0 .8mg· L- 1 )、 Insulin (0 .5～ 6 .0 m g· L- 1 )及 T3(0 .1～ 2 .0μg· L- 1 )对 KCs的增殖均具有不同程度的促进作用。结论 :采用无滋养层法培养大鼠表皮角朊细胞时 ,在培养液中加入一定浓度的EGF、 HGF、 FGF、 CT、 HVB、 Insulin及 T3是必要的。Objective To research the factors influencing keratinocytes (KCs) proliferation activities of rat. Methods The KCs from rats were cultured with non-trophoblast technique in serum medium containing new-born calf serum. Epidermal growth factor(EGF), hepatocyte growth factor(HGF), fibroblast growth factor(FGF), choleratoxin(CT), hydrocortisone(HVB), insulin, and 3,5,3′-triiodothyronine (T 3) were added to the medium at the time of exponential phase of growth. The activities of cell proliferation were detected with MTT method. Results EGF, HGF, FGF, CT, HVB, insulin, and T 3 had promotion effects on KCs proliferation. Conclusion It is benefit for KCs culture to add some factors such as EGF, HGF, FGF, CT, HVB,insulin, and T 3 into medium.
