摘 要:目的 为证实波氏假性霉样菌及尖端赛多孢子菌腹腔局部感染能否引起系统性疾病。方法 采用该菌的不同浓度孢子注射小鼠腹腔 ,记录并统计分析小鼠平均生存时间 (MST)以及该菌在小鼠的肝、脾、肠、肾、肺、脑分离阳性率 ,感染小鼠各脏器病理切片HE及PAS染色。用扫描电镜观察尖端赛多孢子菌临床株在马铃薯培养基(PDA)上的生长物。结果 发现各动物模型组内各菌株组间MST及各脏器菌分离阳性率无差异 ,但不同动物模型组间存在差异。感染小鼠各脏器经病理检查发现孢子、菌丝及中性粒细胞浸润。电镜观察尖端赛多孢子菌临床株可见尖端单生孢子 ,无子囊果 ,无有性阶段。结论 波氏假性霉样菌及尖端赛多孢子菌致病力与是否具有有性阶段无关 ,多次 1× 10 7CFU/ml孢子连续腹腔内攻击免疫抑制小鼠可制备较稳定、可靠的系统性感染小鼠模型 ,并证实了当机体免疫力下降时 ,可由局部感染该菌导致全身性感染性疾病。Objective To certify whether local abdominal infection of Pseudollescheria boydii and Scedosporium apiospermum can induce systemic disease. Methods Spores at different concentrations attacked murine by intraperitoneal inoculation. Mean survival time (MST) of the mice and positive culture result of the fungi isolated from liver, spleen, intestine, kidney, lung and brain in the experimentally infected mice were recorded and statistically calculated. Pathogenic changes of mice were examined by HE stain and PAS stain. A strain of S. apiospermum isolated from a patient was observed by electron scanning microscopy. Results There was no significant difference in MST and the positive rate of the isolated fungi from various organs among three strain subgroups in each model group, but there was statistically significant difference among the different model groups. In those organs of infected animals spores were found with neutrophile exudates and necrotic abscesses. Hyphae were thriven in brain. In S. apiospermum 25018 which was cultured on PDA single spores on the top of conidiogenous cells were seen by electron scanning microscopy. It proved that this strain didn't possess sexual form. Conclusion The virulence of the fungi was not depended on whether it possessed sexual form and by inoculating spores at concentration of 1×10 7 CFU for multiple times, the intraperitoneal challenged murine could be prepared as stable and reliable systemic P. boydii/S. apiospermum infection models. It was concluded that local abdominal infection of P. boydii/S. apiospermum could induce systemic infection when immunosupression occurred.
