清热解毒法对脑出血模型大鼠血SOD、NO、TNF-α的影响

摘　　要：为探讨清热解毒法治疗脑出血的机制 ,6 0只Wistar雄性大鼠以随机表法随机均分为 6组 :假手术 1组、2组 ,模型 1、2组 ,治疗 1、2组 ,对后 4组采用胶原酶诱导大鼠脑出血模型 ,术后 2h治疗 1、2组给予清热解毒中药脑宁康颗粒 (2 4mg·kg- 1·d- 1)灌胃 ,另 4组给予等量生理盐水 ,连续用药 3d(1组 )、7d(2组 )后取血 ,应用荧光分光光度计法和放射免疫分析法测定血SOD、NO、TNF α含量。结果 :与假手术组比较 ,模型组血SOD含量明显下降 (P <0 0 1) ,血NO和TNF α含量均明显升高 (P <0 0 1) ;与模型组比较 ,治疗组血SOD含量明显升高 (P <0 0 1) ,血NO和TNF含量均明显下降 (P <0 0 1,P <0 0 5 ) ;3d、7d组结果相同。结论 :清热解毒法对脑出血后神经细胞损伤具有保护作用 ,其机制可能与抗自由基、抑制NO和TNFTo explore the mechanism of heat-clearing and toxin- relieving herbs in the treatment of cerebral hemorrhage, 60 Wistar male rats w ere randomized into six groups: sham group one, sham group two, model group one, model group two, treatment group one and treatment group two. The rats in the l atter four groups were modeled to be of cerebral hemorrhage by collagenase. Two hours after modeling, treatment groups one and two were given “Nao Ning Kang” at the dosage of 2.4mg/kg -1· d -1, which was effective to clear heat and relieve toxin, treatment group one for consecutive 3 days and group two for 7 days; and the other 4 groups were given equal amount of normal saline. The co ntents of blood SOD, NO and TNF-α were detected by fluorospectrophotometry and radioimmunoassay. Results: In comparison with sham groups, the contents of SOD in model groups fell obviously (P<0.01), while the contents of NO and TNF- α rose obviously (P<0.01); in comparison with model groups, the contents of SOD in treatment groups rose remarkably (P<0.01), while NO and TNF-α fell markedly (P<0.05, P<0.01). It is concluded that heat-clearing and tox in-relieving herbs can protect nerve functions after cerebral hemorrhage possib ly by fighting free radicals and suppressing NO and TNF-α.
