摘　　要：目的 :探讨大鼠急性心肌梗塞 (AMI)后是否存在心肌炎症反应和心脏细胞因子表达。方法 :建立大鼠心梗模型并设假手术组 ,于术后第 1、4、8周末测血流动力学后取心脏。RT PCR检测心脏肿瘤坏死因子 α(TNF α)、白介素 (IL 1β、IL 6、IL 10 )和转化生长因子 β1 (TGF β1 )mRNA的表达 ,并进行心肌病理检查。结果 :与假手术组相比 ,AMI组血流动力学有明显改变。术后第 1周末梗塞区、交界区心肌炎性细胞浸润 ,同期发现梗塞区、交界区和非梗塞区细胞因子表达有不同程度增高 ,其中TGF β1 升高最明显。梗塞后第 4、8周末炎症细胞因子表达下降 ,但TGF β1 仍高表达 ,梗塞区心肌纤维化。结论 :炎症参与心肌梗塞后心室重塑 ,细胞因子在重塑中起一定作用 ,TGF β1 扮演重要角色。Objective:Sought to analyze inflammatory response and the role of the cytokines in ventricular remodeling at different phases after acute myocardial infarction (AMI).Methods:AMI in rats was replicated by ligating the left anterior descending coronary artery.The relative expressions of mRNA for TNF α,IL 1β,IL 6,IL 10 and TGF β 1 in the infarcted,borded and non infarcted myocardium of these rats,as well as in a group of sham operated rats,were assessed by RT PCR.Hemodynamics had been performed before rats were sacrificed.Myocardium was detected by HE staining.Results:There were remarkable differences in hemodynamics between AMI groups and sham groups.Leucocytes recruited in the infracted and borded myocardium at the end of 1 week after AMI.Compared to sham operated groups,expressions of cytokines,such as TNF α,IL 1β,IL 6,TGF β 1 and IL 10 mRNA,were elevated with different extent in infarcted,borded and noninfarcted areas at 1 week after AMI.Expression of TGF β 1 mRNA(4 418 folds) was the highest one.TGF β 1 mRNA expression increased in infarcted and noninfarcted areas at the ent of 4 weeks and 8 weeks after AMI,while other pro inflammatory cytokines decreased.Conclusion:Inflammation and pro inflammatory cytokines take part in ventricular remodeling after AMI.TGF β 1 maybe play a vital role in these pathogenesis after AMI.
