二步酶法测定甘油三酯的自动分析

摘　　要：本文报告四种常用自动分析仪(IL Monarch-1000,Technicon RA-1000,Cobas-MIRA,和Cobas-FARAII)采用二步酶法测定血清甘油三酯(TG)的操作方法.四种分析仪均能有效地排除至少6mmol/L 游离甘油(FG)的干扰。它们的批内CV 在0.66%～1.57%之间;与手工操作的结果比较均无显著差异,t 试验p 均>0.05,且相关良好,r 为0.9957～0.9988.This paper discribes a GPO-PAP two-step method,not affected by free glycerol,for measuring serumtriglycerides by using the automatic analyzers.The free glycerol in sample was removed from the reaction mixtureby pre-incubation with reagent Ⅱ(containing GK,GPO,POD,ATP and 4-CP).The subsquent addition ofreagent Ⅱ(containing lipas and 4-aminoantipyrine)resulted in the formation of color.The efficiency of eliminating free glyceral was fine for all four analyzers,at least 6.0 mmol/L free glycerolcould be removed during the pre-incubationg.The coefficient of variation of within-run were<1.0% forCobas-Mira and Fara Ⅱ,<1.5% for Technicon RA-1000 and<1.6% for IL Monarch-1000.The resultlinearity ranges were 12.0,11.0,and 8.0 mmol/L for IL Monarch-1000 and Cobas-Mira,Cobas-Fara Ⅱand Technicon RA-1000,respectively.The results estimated by four analyzers corralated closely with those ob-tained by manul operation and the correlation coefficients between both assays were 0.995-0.9988.
