摘 要:目的 研究槲皮素、芦丁及葛根素等抑制 HDL 氧化修饰的作用。方法 按一次性密度梯度超速离心法分离人血清 HDL,分别加 5 μmol/ L 槲皮素、芦丁及葛根素与 HDL 温育 3h,然后利用 Cu2 +介导体外氧化修饰脂蛋白的方法 ,观察 HDL 氧化修饰不同时间 ,其 A2 34 、REM、TBARS及蛋白质羰基的动力学改变 ,并与不加药物的对照组比较。结果 Cu2 + 介导氧化 HDL修饰时 ,在 2、4、6、8、12及 2 4 h后 ,其 A2 34 、REM、TBARS及蛋白质羰基含量均逐步增加 ;加入 5 .0 μmol/ L 槲皮素、芦丁及葛根素的实验组 ,Cu2 + 介导 HDL 的氧化修饰分别延迟 2~ 4 h、4 h和2~ 4 h,其 A2 34 、REM、TNAR及载脂蛋白羰基含量较对照组降低 ;槲皮素实验组分别降低 2 4 .2 %~ 4 9.0 %、3.0 %~ 2 5 .0 %、7.9%~ 34.1%及 2 8.3%~ 5 0 .4 % ;芦丁实验组降低 9.4 %~ 4 2 .0 %、4 .0 %~ 2 0 .7%、1.2 %~2 6 .8%及 18.6 %~ 6 4 .6 % ;葛根素实验组载脂蛋白羰基含量较对照组降低 14 .1%~ 5 2 .1% ,而其 A2 34 、REM,TBARS虽有降低 ,但降低幅度不大。结论 槲皮素及芦丁可显著抑制 HDL 的氧化修饰 ,而葛根素的抑制作用较小。Objective To evaluate the inhibitory effect of quercetin, rutin and puerarin on the HDL oxidation induced by Cu 2+ and to investigate their action on the prevention and cure of atherosclerosis. Methods The serum HDL of healthy human was isolated by the one step density gradient ultracentrifugation. The HDL oxidation was induced by Cu 2+ in vitro for different time. quercetin and rutin at 5 μmol/L were added ahead, respectively. HDL was oxidized, then identified by A 234, REM, TBARS and protein carbonyls content, and was compared with control group. Inhibition of puerarin was weak. Results ① A 234, REM, TBARS and protein carbonyls formation increased gradually during HDL oxidation induced by Cu 2+ in vitro. ② During HDL oxidation induced by Cu 2+ in vitro and incubation with quercetin and rutin, the kinetic changes of A 234, REM, TBARS and protein carbonyls formation showed lag phases of 2 h,2-4 h,2-4 h and 4 h respectively, and showed reductions by 24.2%-49.0%(P<0.001) and 9.4%-42.0%(P<0.01); 3.0%-25.0% and 4.0%-20.7%(P<0.05); 7.9%-34 1% and 1.2%-26.8%(P<0.05);28.3%-50.4% and 18.6%-64.6% (P<0.001) respectively. In the puerarin group, only protein carbonyls formation showed a lag phase of 4 h and a reduction by 14.1%-52.1%(P<0.001) while its A 234, REM, TBARS reduced to a small extent. Conclusion The above findings suggested that quercetin and rutin inhibit oxidation of HDL significantly, but puerarin has less antioxidative function.
