摘  要:目的 :实验观察苦参碱口服给药对化疗诱导的多药耐药基因表达产物P170 、肺耐药蛋白LRP和拓扑异构酶Ⅱ的影响 ,探讨其干预多药耐药的分子生物学基础 ,以指导临床应用苦参碱干预化疗多药耐药的产生。方法 :以亚于治疗剂量的联合化疗PFC方案 ,建立小鼠S180 肿瘤细胞多药耐药模型 ,同时给予苦参碱 4周 ,流式细胞仪荧光检测P170 、LRP、TOPOⅡ。结果 :苦参碱明显降低化疗诱导后耐药细胞P170 、LRP的表达率和ROPOⅡ活性。说明苦参碱可通过对相关生物活性物质的调节 ,干预化疗诱发的肿瘤细胞多药耐药的产生。Objective: To observe the effect of matrine on P 170 expressed by multi-drug resistance gene, lung resistant protein(LRP), and boposomeras Ⅱ. Methods: By the method of less dosage of chemotherapy PFC, the mouse model of multi-drug resistance of S 180 tumour cell was set up. At the same time, matrine was administered to the mouse model for 4 weeks, and then P 170 , LRP, TOPOⅡ were observed by flow cytometry. Results: Martine can obviously reduce the express of P 170 , LRP and the activity of TOPOⅡ in multi-drug resistance tumour cell induced by chemotherapy. Conclusion: Matrine can intervene the occurrence of the multi-drug resitance of tumor cells induced by chemotherapy.
