姜黄素对膀胱癌EJ细胞的作用研究

摘　　要：目的 :观察姜黄素对膀胱癌系细胞EJ的体外作用 ,并探讨其可能的作用机制。方法 :应用光镜形态学 ,MTT法、流式细胞仪和免疫细胞化学法观察了 5、10、2 0mg/L姜黄素在体外对膀胱癌EJ细胞的生长抑制和诱导凋亡作用 ,以及对细胞DNA含量 ,对NF κB、CyclinD1表达的影响。结果 :所有浓度组姜黄素均对膀胱癌EJ细胞有明显的生长抑制作用 ,抑制率≥ ( 2 7 5± 3 1) % (P <0 0 5 )。 10mg/L以上浓度均可出现显著的诱导调亡作用 ,凋亡率≥ 4 6 %± 1 8% (P <0 0 5 ) ,下调NF κΒ(表达率≤ 35 8%± 4 2 % ,P <0 0 5 ) ,下调CyclinD1的表达 (表达率≤ 2 9 7%± 3 2 % ,P <0 0 5 )。姜黄素导致的细胞周期阻滞以G1期为主 ,使S期细胞比例显著减少 (P <0 0 5 )。结论 :姜黄素对膀胱癌细胞系EJ有明显的生长抑制和诱导凋亡作用 ,其作用机制与其影响细胞周期和下调NF κB、CyclinD1的表达相关 。Objective:To observe the effect of curcumin on bladder cancer cell line EJ in vitro.Methods:Cell morphology,MTT,flow cytometer,immunocytochemical method for detecting NF-κB,Cyclin D 1 were used to observe the effect of 5,10,20 mg/L curcumin on bladder cancer cell line EJ in vitro.Results:All concentrations curcumin resulted in the growth suppression significantly [Suppression ratio ≥(27.5±3.1)%,P<0.05].Above 10 mg/L concentrations curcumin induced apoptosis [Apoptosis ratio≥(14.6±1.8)%,P<0.05] and down-regulated of the expression of NF-κB[Expression ratio≤(35.8±4.2)%,P<0.05],Cyclin D 1[Expression ratio≤(29.7±3.2)%,P<0.05].The cell phase arrest induced by curcumin was G 1 phase arrest mainly with significant decrease of S phase.Conclusions:Curcumin can suppress the growth,induce apoptosis of bladder cancer EJ cell in vitro.Its mechanism is related with down-regulations of the expressions of NF-κB and Cyclin D 1.Curcumin has great potential for the treatment of bladder cancer.
