摘　　要：依据单纯疱疹病毒（ＨＳＶ）ＤＮＡ多聚酶基因的保守序列设计其型公共引物，采用二步温控法，成功地建立起ＰＣＲ反应体系。将４０份拟诊为病毒性脑炎患者的脑脊液（ＣＳＦ）及多种对照标本进行ＨＳＶ的ＰＣＲ扩增分析，并用ＰＣＲ制备生物素标记的ＤＮＡ探针与ＰＣＲ检测产物进行斑点杂交。结果表明，８份ＣＳＦ标本可检出ＨＳＶ－ＤＮＡ。单纯疱疹性脑炎占总数的２０％。该ＰＣＲ可作为单纯疱疹性脑炎的早期诊断手段。A pair of general primers was selected according to a conservative segment of herpes simplex virus(HSV) DNA polymerase gene,and tow-protocal polymerase chain reaction was established to detect the HSV-DNA in cerebrospinal fluid samples of patients with suspected viral encephalitis and other control samples. The products was assayed by electrophoresis and blot hybridization with biotin-labelled HSV-DNA probes which were generated by PCR. The results demonstrated that the tow-protocal PCR is successful with a excellent specificity and high sensitivity.This PCR procedure is simple and worth to popularize as an early diag nostic method for HSE.The positive rate of HSV-DNA in 40 patients with suspected viral encephalitis was 20%(8/40).The patients with HSE showed some character of findings by manifestation and clinical assay.
