人降钙素(hCT)放免试剂盒的研制及其临床应用

摘  要:作者在人血清降钙素的放免试剂盒研制过程中,hCT的^(125)I标记采用Iodogen法,以Sep—PakC_(18)柱分离纯化,通过优选建立了hCT放免非平衡竞争结合分析法。通过方法学和可靠性鉴定,说明了该方法具有较高的特异性、精密度、准确度和灵敏度,在八周内观察,具较好的稳定性。100例正常人测定hCT水平为74.7±30.5ng/L,而75例肾炎、55例肺癌、34例乳腺癌和5例甲状腺髓样癌患者其检测值分别为385.3±159.6ng/L,206.5±77.3ng/L,157.3±65.2ng/L,579.5土357.5ng/L,均明显高于正常对照组(p<0.01)。经临床验证,该试剂盒符合稳定、可靠、灵敏、适用的要求。We report the preparation of a RIA kit for human calcitonin. Human calci-toinn (hCT) was labelled with iodogen method and then purified withSep-Pak l8 column.High specificity, precision, accwroky and cesisitivity were noted in methodological sludy with reliable stability at eight weeks. The serum hCT value was 74.7±30.5ng/L (x±S) in 100 healthy subjects, In patients with chronic renal fai!ure(75 cases), lung cancer (55cases),breast career (34 cases), and medullary thyroid carcinoma (5cases), the values were 385±159.6ng/L, 206.5±77.3ng/L, 157.3±65.2ng/L, 579.5±357.5ng/L respectively. All of these were significantly higher than those of the healthy group (p<0.01). The kit is found to be clinically applicable.
