摘 要:为了进一步分析稀土元素镧化合物——硝酸镧(La(NO_3)_3)在不同浓度和不同作用条件下对培养的单个大鼠平滑肌细胞内重要元素的影响,我们继续应用同步辐射X射线荧光技术分析10^(-4)mol/L的La(NO_3)_3对单个细胞内重要元素水平的影响。实验观察到10^(-4)mol/L La(NO_3)_3即刻和持续作用均能使单个平滑肌细胞内La显著升高,与此同时Ca、P、S也发生高度相关性的变化,K、Fe、Cu、Zn、Ni等元素含量也有不同程度的改变。实验结果提示:La^(3+)进入细胞内并影响细胞内某些重要元素的分布,这可能是La^(3+)引起细胞增殖和细胞毒性的机制之一。To further understand the effect of La(NO_3)_3 on smooth muscle cell of rat for different doses and different action time, the synchrotron radiation X-ray fluorescence(SRXRF) micro-probe was used to determine the level of essential elements including P, S, K, Ca, Fe, Cu, Zn, Ni and La in single smooth muscle cell(SMC). The change of element levels in SMC upon application of 10^(-4)mol/L La(NO_3)_3 was observed. La was hardly detected in SMC before application of La(NO_3)_3. After application of 10^(-4)mol/L La(NO_3)_3, the levels of La inside cell increased significantly and so did Ca. The levels of Ca, P, S appear to be highly correlated with the level of La in single SMC. These results suggest that La^(3+) could be taken up by SMC, which in turn induces change of level of intracellular essential elements. This maybe one of the machanisms by wich La^(3+) causes in cell proliferation and toxicity.
