中药含药血清对AngⅡ诱导的心肌细胞原癌基因c-fos表达的影响

摘　　要：目的 :探讨中药开心胶囊抗心肌肥大的作用机制。方法 :通过分离、培养乳鼠心肌细胞 ,给予不同剂量开心胶囊和开搏通干预 ,采用血管紧张素 ( Ang )诱导心肌细胞原癌基因 c fos表达 ,用逆转录聚合酶链反应 ( RT PCR)法检测心肌细胞 c fos表达。结果 :不同剂量开心胶囊含药血清与开搏通一样均具有抑制心肌细胞 c fos表达的作用 ( P均 <0 .0 5 )。结论Objective: To explore the mechanism of traditional Chinese medicine Kaixin capsule(开心 胶囊) on antimyocyte hypertrophy. Methods: Cardiac myocytes in vitro isolated from neonatal rats were cultured and were given different dosage of Kaixin capsule and captopril. Angiotensin Ⅱ was used to induce the expression of protooncogene cfos in cardiac myocytes. Expression of protooncogene cfos was measured by the method of reverse transcriptionpolymerase chain reaction(RTPCR). Results: Pretreatment with Kaixin capsule absorption serum had the effect of inhibiting the expression of protooncogene cfos as same as captopril(all P <0 05). Conclusion: Kaixin capsule possesses the effect of inhibiting cardiac myocyte hypertrophy.
