摘 要:目的 :观察小青龙汤对哮喘模型小鼠肺组织内 Th1/ Th2比值的影响并探讨其作用机制。方法 :将4 9只雌性 BAL B/ c小鼠随机分为正常对照组 ( A组 )、哮喘组 ( B组 )、小青龙汤小剂量组 ( C组 )、小青龙汤大剂量组 ( D组 )、地塞米松组 ( E组 )、必可酮组 ( F组 )及联合用药组 (减半的小剂量小青龙汤加地塞米松 ,G组 ) ,每组 7只。卵蛋白致敏复制哮喘模型 ,激发哮喘前 1h,各组小鼠分别使用相应药物治疗。激发 12 d后断头处死小鼠 ,立即取肺脏进行免疫组化染色 ,观察组织中 Th1和 Th2表达。结果 :1各组哮喘小鼠肺组织内 Th1和 Th2数量均显著增多 ( P均 <0 .0 1) ;D组较 C组显著减少 ( P <0 .0 5 ) ;D、E、F、G组之间差异均无显著性 ( P均 >0 .0 5 )。 2 B组小鼠肺组织内 Th1/ Th2比值非常显著性降低 ( P<0 .0 1) ,肺内细胞因子呈明显的 Th2表型变化 ;各治疗组较 B组均呈非常显著性升高 ( P均 <0 .0 1) ,肺内细胞因子呈明显的 Th1表型变化 ;各治疗组间差异均无显著性 ( P均 >0 .0 5 ) ,且接近 A组。结论 :1哮喘小鼠肺组织内 Th1/ Th2比值明显降低 ,支持哮喘发病中存在 Th1/ Th2比值失衡学说。 2小青龙汤选择性降低肺组织内 Th1、Th2数量 ,逆转失衡的 Th1/ Th2比值是其治疗哮喘的重要机制之一。Objective: To investigate the effects of Xiaoqinglong decoction(小青龙汤) on Th1/Th2 balance in the mice models of asthma and explore its mechanism. Methods: Fortynine female BALB/c mice were randomly divided into control group (group A), asthma group (group B), small dose of Xiaoqinglong decoction group (group C), large dose of Xiaoqinglong decoction group (group D), dexamethasone group (group E), beclomethasone group (group F), combination drugs group (group G), 7 mice were in each group. The mice models of asthma were replicated by egg albumin hypersensitivity. At 1 hour before being challenged each day, the mice in group C were administered with small dose of Xiaoqinglong decoction, so were group D with large dose of Xiaoqinglong decoction, group E with dexamethasone, group F with beclomethasone , group G with half dose of small dose of Xiaoqinglong decoction and dexamethasone. Twelve days after being challenged, 7 mice in each group were executed, their lungs were sampled immediately and stained by immunohistochemistry dye to observe the expression of Th1 and Th2. Results: ①The numbers of Th1 and Th2 in the lungs of all the asthma mice were significantly higher(all P <0 01), they were lower in group D than those in group C(both P <0 05). ②The differences of the numbers of Th1 and Th2 in the lungs were not significant among group D, E, F and G(all P >0 05). The value of Th1/Th2 in the lungs of group B was significantly decreased( P <0 01), and the cytokines expression in the lungs was turned into Th2. The value of Th1/Th2 in the lungs of each treatment group was very significantly higher than that of group B respectively (all P <0 01), and the cytokines expression in the lungs was turned into Th1. The difference of the value of Th1/Th2 in the lungs wasn't significant among each treatment group,they were closely to the levels of group A(all P >0 05).
