高同型半胱氨酸血症促进白介素-8的表达及辛伐他汀的拮抗作用

摘　　要：目的　研究高同型半胱氨酸血症 (Hhcy)对白介素 8(IL 8)的表达和NF κB活化的影响 ,以及辛伐他汀对此的拮抗作用。方法　 2 1只雄性新西兰兔随机分成 3组 ,对照组 (正常饮食 ) ,蛋氨酸组 (高蛋氨酸饮食 ) ,辛伐他汀组(高蛋氨酸饮食 +辛伐他汀 ) ,12周后 ,测定血浆同型半胱氨酸 (Hcy) ,ELISA方法检测IL 8蛋白水平 ,免疫组化检测血管NF κB表达和活化。结果　高蛋氨酸饮食显著增加了血浆Hcy水平 ,蛋氨酸组IL 8的产生增加 ,主动脉内皮细胞NF κB表达增强。辛伐他汀降低IL 8水平 ,减少了NF κB的表达和活化。结论　Hhcy诱导NF κB的活化 ,从而促进IL 8的表达 。Objective To investigate the effect of hyperhomocysteinemia on NF-κB mediated IL-8 production and possible antagonizing effect of simvastatin.Method Twenty one New Zealand white rabbits were ramdomly divided into three groups ,control(C,normal feeding),methionine(M 1.7%methionine)and simvastatin(S 1.7%methionine and simvastatin).The level of Hcy was measured ,IL-8 was determined by ELISA ,the expression and activation of NF-κB were studied by immunohistochemistry after twelve weeks.Results Hyperhomocysteinemia was induced after high methionine diet,the production of IL-8 and activation of NF-κB were increased by hyperhomocysteinemia.Simvastatin inhibited Hcy-induced production of IL-8 and activation of NF-κB.Conclusion Hyperhomocysteinemia induces activation of NF-κB and consequently enhances the production of IL-8,while simvastatin can antagonize such stimulative effect.
