兔眼玻璃体腔地塞米松磷酸钠的高效液相色谱法测定

摘　　要：目的 :建立高效液相色谱法测定新西兰大白兔眼玻璃体地塞米松磷酸钠含量。方法 :玻璃体溶液 0 .1mL经 70℃干燥变性蛋白并浓缩标本 ,流动相 2 0 μL溶解浓缩回收 ,离心。色谱条件为流动相 :甲醇 0 .34%磷酸二氢钾 (5 5∶4 5 ) ,流速 :0 .2 0mL·min-1,C18色谱柱 ,柱温 :2 5℃ ,检测波长 :2 4 0nm。结果 :地塞米松磷酸钠在 0 .0 6 2 5～ 2 .5mg·L-1内线性关系良好(r =0 .9988) ,方法回收率为 10 0 .2 %～ 10 6 .3% ,绝对回收率为 70 .7%～ 80 .1% ,日内、日间RSD分别为 2 .9%～ 4 .4 % ,5 .1%～ 8.3% ;检测灵敏度 0 .0 2 0mg·L-1。结论 :标本前处理可以有效去除蛋白 ,过程简单 ,方法稳定 ,灵敏度高 ,结果可靠 。OBJECTIVE To establish a method for determination of dexamethasone sodium phosphate in vitreous body of New Zealand rabbit by HPLC.METHODS Procedures of sample included deproteinization and condensation at 70 ℃ ,and recovery by mobile phase.Mobile phase of methanol 0.34% KH 2PO 4(55∶45) and ZOBARX C 18 column were used,the flow rate was 0.20 mL ·min -1 and detection wavelength 240 nm .RESULTS The linear range of dexamethasone sodium phosphate was 0.062 5 - 2.5 mg ·L -1 ( r = 0.998 8 ).The absolute and relative recoveries of three levels were 100.2% - 106.3% 、 70.7% - 80.1% .The RSD of intra day and inter day were 2.9% - 4.4% 、 5.1% - 8.3% respectively.The limit of detection was 0.020 mg ·L -1 .CONCLUSION This method has been proved to be accurate and reliable.
