绒螯蟹凝集素的红细胞凝集活性分析

摘  要:利用血凝试验及糖抑制试验测定一种绒螯蟹的血清及肌肉提取液的凝血活性及糖抑制专一性,同时进行热稳定性、pH 及 Ca 影响试验。结果显示:血清只能凝集 9 种红细胞,肌肉提取液能凝集 12 种红 2+细胞。血清对鹌鹑红细胞的凝集可被乳糖、D-甘露糖、D-果糖、D-半乳糖 4 种糖所抑制,最小抑制浓度均为 200 mmol/L,其凝集活性均依赖于 Ca ,在 pH 为 6.0~7.0 范围内较稳定,经 50 ℃ 保温 10 min 后活性 2+完全消失;肌肉提取液只被 D-甘露糖抑制,在 pH 为 4.0~10.0、温度为 4 ℃~90 ℃ 范围内均具活性,说明有很强的耐热性,其凝集活性不依赖于 Ca 。以上结果表明:血清与肌肉提取液均含有凝集素,且凝集 2+素的种类不同。Serum and muscular extract from Eriocheir sp. were detected for hemagglutinating activity, glyco-inhibiting action. Besides, thermostability and influence of pH and Ca2+ to the activity of them were tested. The results showed that serum could only agglutinate nine erythrocytes, and muscular extract could agglutinate twelve erythrocytes. Agglutinative activity of serum to quail erythrocytes was inhibited by lactose, D-mannose, D-fructose and D-galactose, and the minimum inhibitory concentration was 200 mmol/L. Serum hemagglutination activity depends on Ca2+, and it is stable over pH 6.0-7.0 and lost at 50 ℃ by 10 minutes; muscular extract agglutination quail erythrocytes were only inhibited by D-mannose, it had agglutinating activity in the pH range of 4.0- 10.0, and the temperature range of 4℃-90℃and their hemagglatination activity doesn't depend on Ca2+. The results indicated that both of serum and muscle of Eriocheir sp. contain agglatinin, and they are different in nature.
