摘  要:为观察NGF等对小鼠骨髓基质细胞的诱导分化作用,从小鼠骨髓中分离培养基质细胞并传至第3代,采用bFGF预诱导24 h,NGF、维甲酸、β-巯基乙醇等诱导24 h后,进行NF-200、GFAP、fibronectin免疫荧光染色,荧光显微镜观察免疫阳性细胞。结果表明:诱导后小鼠骨髓基质细胞形态改变不明显,但出现表达NF-200、GFAP的细胞,而表达fibronectin的细胞减少,对照组几乎无NF-200和GFAP免疫阳性细胞,主要为表达fibronectin细胞。结论:NGF等可以在体外诱导小鼠骨髓基质细胞分化成表达NF-200和GFAP细胞。To investigate the inducing differentiation with NGF (nerve growth factor) on mouse bone marrow stromal cells (mBMSCs) in vitro, the mBMSCs from mouse were isolated and cultured for 3 passages, which were induced by NGF and all-trans-retinoic acid (RA) and β-mercaptoethanol for 24 h after pre-induced by basic fibroblast growth factor (bFGF) for 24 h. And the cells were stained immunofluorescently with neurofilament-200 (NF-200) and glial fibrillary acidic protein (GFAP) and fibronectin antibodies. After induction many cells expressed NF-200 and GFAP. The numbers of fibronectin positive cells decreased. However, in control group, the cells expressed mainly fibronectin without NF-200 and GFAP positive cells. The above results demonstrate that the mBMSCs can be induced to differentiate into NF-200 and GFAP expressed-cells by NGF.
