脂质体阿霉素治疗SRS淋巴瘤小鼠的心脏病理变化

摘　　要：将阿霉素包封在多层脂质体中,与游离阿霉素比较,观察治疗SRS淋巴瘤小鼠过程中小鼠心脏的病理改变。当剂量均为15mg/kg时,光镜见游离阿霉素治疗组小鼠心肌组织单核细胞浸润,肌细胞肿胀,广泛空泡变性,易见细胞坏死;而脂质体阿霉素组仅见心肌细胞轻度肿胀,少见空泡变性,未见坏死。电镜观察到游离阿霉素组小鼠心肌细胞线粒体肿胀、破裂,肌丝断裂、溶解。而脂质体阿霉素组仅见局灶性肌丝片段丢失。结果表明,脂质体载药能明显降低阿霉素的心脏毒性。Adriamycin was capsulated in multivesicular liposomes composed of lipids of egg origin. Light microscopic examination of cardiac sections taken from SRS lymphoma mice receiving 15mg/kg free adriamycin revealed noticeable histologic changes on day 3,and by day 7 the cardiac damage became severe.Cellular edema,monocytic infiltration,and cell necrosis were evident.In contrast,sections taken from mice receiving 15mg/kg liposomal adriamycin domonstrated only slight edema and no cell necrosis was found by day 7.Ultrastructurally,the loss of cytoplasmic elements,swelling and distortion of mitochondria,extensive vacuolization and iysis of myofila- ment were noted on mice treated by free adriamycin.Focal cellular dilatation and fragmental loss of myofilament were observed on mice treated by liposomal adriamycin.The results showed that the cardiotoxicity of adriamycin on SRS lymphoma mice using liposome as drug carrier decreased remarkedly.
