单克隆抗体用于比较禽呼肠孤病毒株的抗原关系  

COMPARISON OF ANTIGEN RELATIONSHIP OF AVIAN REOVIRUS WITH MONOCLONAL ANTIBODIES

摘　　要：利用淋巴细胞杂交瘤技术在国内首次建立了8株分泌禽呼肠孤病毒(ARVS)的单克隆抗体(McAb)杂交瘤细胞株,腹水效价在10^(-3)～10^(-5)之间。用琼脂扩散试验鉴定,5株属 IgGl,2株属IgG2b,1株属 IgG3.ELISA 阻断试验表明,4个 ARVS 分离株的抗原结构明显不同,提示利用 McAb 进行 ARVS 的分型具有很大潜力.在与两个国内毒株(广州分离株和 NAU-8902)分别发生特异性反应的4株 McAb 中,有3株是相同的,说明两者具有很近的亲缘关系.试验还发现,McAb D19株能被试验的所有毒株阻断,提示 D19株有希望成为检测 ARVS 的群特异性 McAb。Eight strains of hybridoma cell lines secreting McAb against avian reovirus(ARVS)were es-tablished with lymphocyte hybridoma technique.The ELISA titres of McAb from mice ascites fluid were10^(-3).to 10^(-5) Five strains of McAb belong to the subclass IgG1,2 to IgG2b and 1 to IgG3,as identifiedwith immunodiffusion test.The antigen relationship among 4 isolates of ARVS in ELISA-blocking test re-vealed significant difference in antigenic structure.This indicated that the McAb might be useful in compari-son of antigen relationship of ARVS and in typing of ARVS serotypes.Of 4 McAb strains reacting with 2 na-tive isolates(strains Guangzhou and NAU-8902),3 were identical,which suggested a closely relationshipamong them.In addition,McAb D19 can be blocked by all ARVS isolates tested and therefore might beused as a common McAb in survey of ARVS.
