摘  要:目的:比较多种药物对体外培养皮肤组织中细胞因子表达的影响。方法:咪唑斯汀、氯雷他定、西替利嗪、非索非那定、地塞米松、环孢素等药物和皮肤组织体外共培养,同时加入组胺、花生四烯酸等致炎刺激物,培养24h后逆转录PCR法测定上述药物作用后皮肤组织中单核细胞趋化蛋白(MCP)-1、MCP-3、活化后可调节的正常T细胞表达分泌的因子(RANTES)和嗜酸性粒细胞趋化因子eotaxin等的mRNA表达;ELISA法测定培养上清液中细胞因子白介素(IL)-4、IL-5和肿瘤坏死因子(TNF)-α等炎症递质的含量。结果:咪唑斯汀组中MCP-1基因的表达显著低于西替利嗪、氯雷他定、非索非那定组;MCP-3基因的表达显著低于西替利嗪、非索非那定组;eotaxin和RANTES的表达显著低于氯雷他定组(P<0.05);培养上清液中的IL-4含量显著低于非索非那定组,IL-5的含量显著低于西替利嗪、氯雷他定、非索非那定组,TNF-α的含量显著低于氯雷他定组(P<0.05)。结论:比较不同的抗组胺药,咪唑斯汀显示有较强的抗炎症作用。Objective: To investigate the inhibitory effects of different antihistamines on the expression of chemokine of cultivated skin in vitro. Methods: The skin was cultivated with mizolastine, cetirzine, loratadine, fexofenadine, dexamethasone and cyclosporin A, respectively. Meanwhile histamine and arachidonic acid were added. Twenty-four hours later the total RNAs were obtained and purified with tri-pure isolation reagent. The mRNA expression levels of monocyte chemotactic protein-1(MCP-1)?monocyte chemotactic protein-3(MCP-3)?regulated on activation, normal T cell expressed and secreted(RANTES)and eotaxin were analyzed by RT-PCR methods. The concentrations of interlukin(IL)-4?IL-5 and tumor necrosis factor-α(TNF-α) in cultivated supernatant were detected by ELISA. Results: In the mizolastine group the MCP-1 mRNA expression was significantly lower than that in cetirzine,loratadine or fexofenadine group; the MCP-3 mRNA expression was significantly lower than that in cetirzine or loratadine group; the RANTES mRNA expression was significantly lower than that in loratadine group; the eotaxin mRNA expression was significantly lower compared with that in loratadine group(P < 0.05). The level of IL-4 in supernatant in mizolastine group was significantly lower than that in fexofenadine group, and the level of IL-5 was significantly lower than that in cetirzine ,loratadine and fexofenadine groups, while the level of TNF-α was significantly lower than that in loratadine group. Conclusion: Mizolastine can inhibit the expression of chemokines and cytokines more strongly than the other antihistamines.
